BlaPSZ-1, a novel AmpC gene identified from a Pantoea isolate

چکیده

Background Pantoea species of the family Erwiniaceae are well-known plant pathogens and animal human conditional pathogens. Due to widespread continuous use antimicrobials, multidrug-resistant strains continue emerge, making clinical treatment difficult; therefore, there is an increasing need clarify mechanisms drug resistance. Methods A rabbit anal fecal sample was collected by a swab streak plate method used isolate single colonies. The standard agar dilution determine minimum inhibitory concentrations (MICs) against antimicrobials. complete genome sequence bacterium obtained using Next-Generation Sequencing platforms. potential resistance gene annotated based on Comprehensive Antibiotic Resistance Database (CARD) verified molecular cloning. β-lactamase PSZ-1 expressed via pCold I expression vector its enzyme kinetic parameters were analyzed. genetic environment evolutionary process novel gene-related sequences analyzed bioinformatic methods. Results endophytica X85 showed some degree penicillins as well cephalosporins. AmpC gene, designated bla in this research, identified be encoded plasmid (pPEX85) P. X85. Bla several first-, second-and third-generation cephalosporins aztreonam, but it did not show fourth-generation or carbapenems tested. Enzyme assays revealed that could hydrolyze amoxicillin, penicillin G, cephalothin, cefazolin, hydrolytic activity strongly inhibited inhibitor avibactam, which generally consistent with antimicrobial susceptibility testing results. No observed for aztreonam. Conclusion In study, PSZ-1, characterized It shows discovery can help guide scientific drugs husbandry practice, effectively avoiding abuse antimicrobials thus preventing further development spread bacterial

Characterization of a novel AmpC β-lactamase produced by a carbapenem-resistant Cedecea davisae clinical isolate.

A Cedecea davisae isolate, which was intermediate or resistant to third-generation cephalosporins and carbapenems, was recovered from a urine sample. Susceptibility testing, isoelectric focusing, and analysis of outer membrane proteins showed that AmpC β-lactamase expression combined with porin deficiency accounted for the carbapenem resistance. A cloning experiment followed by phenotypic and e...

Analysis of a novel gene and beta-galactosidase isozyme from a psychrotrophic Arthrobacter isolate.

We have characterized a new psychrotrophic Arthrobacter isolate which produces beta-galactosidase isozymes. When DNA from this isolate was transformed into an Escherichia coli host, we obtained three different fragments, designated 12, 14, and 15, each encoding a different beta-galactosidase isozyme. The beta-galactosidase produced from fragment 12 was of special interest because the protein su...

Molecular Characterization of a Salmonella Typhimurium Isolate from Caspian Pony

Typhoid disease or salmonellosis is a common sickness in horses. In several epidemiological studies inhospitalized horses, several serotypes of Salmonella often are predominant in nosocomial infections.Transportation, overcrowding, dehydration, oral antimicrobial therapy and infections are the risk factorswhich may activate latent or subclinical salmonellosis. In this study, t...

CTL Responses to a DNA Vaccine Encod-ing E7 Gene of Human Papillomavirus Type 16 from an Iranian Isolate

Background: Cervical cancer is the most prevalent tumor in developing countries and the second most frequent cancer among female population worldwide. Specific human papillomaviruses and, most notably, HPV types 16 and 18 are recognized as being caus-ally associated with cervical carcinomas. The early HPV type 16 genes, E6 and E7, di-rectly participate in the in vitro transformation of primary ...
